SG-094 (SG094) is a specific cytostatic two-pore channel 2 (TPC2) inhibitor with IC50 of 8.3 uM (TPC2 activator TPC2-A1-P inhibition), shows antiproliferative effect on RIL175 cells with IC50 of 3.7 uM.
SG-094 blocked PI(3,5)P2-elicited TPC2 currents, significantly increased the percentage of channel inhibition (75%) compared with tetrandrine.
SG-094 had no considerable inhibitory effect on TRPML1, only on TPC1.
SG-094 inhibits TPC2 upon activation with both TPC2-A1-P and and TPC2-A1-N.
SG-094 significantly reduced VEGF-induced phosphorylation of several downstream targetseNOS, JNK, and AKT in cells, without decreasing their total protein.
SG-094 influenced cellular energy metabolism and was less toxic than tetrandrine to non-cancerous cells, reduced tumor growth in vivo.
