SETD8 inhibitor C23 is a novel chemical inhibitor of SETD8, inhibits H4K20 methylation, inhibits IR-induced 53BP1 foci.
C23 does not affect the formation of foci for phosphorylated H2AX (γH2AX) in U2OS cells.
C23 phenocopies the impact of the SETD8 inhibitor UCN0379 in DNA replication.
C23 inhibits 53BP1 recruitment to DSBs and 53BP1-dependent repair activities.
