Dalzanemdor (SAGE-718) is a potent and high intrinsic activity N-Methyl-d-aspartate receptor (NMDAR) positive allosteric modulator with EC50 of 86 and 79 nM for GluN2A and GluN2B, respectively, reverses DOI-induced increases in locomotor activity.
Administration of SAGE-718 (1 mg/kg DOI) significantly increased locomotor activity and rearing compared to vehicle.
Acute administration of SAGE-718 (5-10 mg/kg) significantly reduced DOI-induced increases in distance traveled and rearing during the first 30 min after DOI administration.
SAGE-718 is currently in a placebo-controlled phase 2 clinical study to evaluate its impact on cognitive deficits in an HD patient population.
