Ryanozole (TMDJ-035) is the first highly potent and selective ryanodine receptor 2 (RyR2) inhibitor with EC50 of 13 nM, shows no effect on RyR1 and RyR3.
TMDJ-035 (30-100 nM) is effective in preventing generation of Ca2+ waves in cardiomyocytes without affecting action potential-induced Ca2+ transients.
TMDJ-035 is a promising candidate for drug development to treat potentially fatal RyR2-related cardiac arrhythmias.
