RA-0002034 is a covalent, specific and cell-active chikungunya virus (CHIKV) nsP2 protease (nsP2pro) inhibitor with IC50 of 58 nM.
RA-0002034 covalently modifies the catalytic cysteine in a site-specific manner.
RA-0002034 shows no significant off-target reactivity against a panel of cysteine proteases.
RA-0002034 effectively inhibits viral replication of both CHIKV and related alphaviruses in cellular models of alphavirus infection.
RA-0002034 inhibits CHIKV replication with an EC50 of 11 nM and VEEV replication with an EC50 of 320 nM, suggesting activity across multiple alphaviruses.
