Pyrcoumin is a competitive inhibitor of dCTP pyrophosphatase 1 (dCTPP1, IC50=3.3 uM), inhibits Wnt signaling with IC50 of 8.4 uM in SuperTOPFlash reporter gene assays.
Pyrcoumin dose-dependently decreased the expression of Wnt target genes AXIN2, CCND1 and LEF1 in Wnt-activated HEK293 cells.
Pyrcoumin inhibited Wnt pathway activity also in HCT116 cells, did not affect the abundance of β-catenin, or other pathway components such as Axin2 and phosphorylated (Ser33/Ser37/Thr41) β-catenin.
Pyrcoumin impairs the direct interaction of dCTPP1 and USP7.
