PVP-037.2 is a small-molecule that induces cytokine production and TLR7/8 agonist, demonstrates TLR7-dependent enhancement of TH1-polarizing adjuvanticity.
ODN 20958 (TLR7 specific antagonist) completely blocks PVP-037.2-induced TNF production in human PBMCs.
PVP-037.2 acts in synergy with a squalene-based oil in water formulation to enhance cytokine induction in vitro and vaccine immunogenicity in vivo.
The type 1 immunomodulatory profile of PVP-037.2 is mediated by TLR7.
Formulation of PVP-037.2 with squalene enhanced its adjuvanticity.
PVP-037.2 demonstrates a TLR7/8-dependent immunomodulatory profile.
