PUS7 inhibitor C17 is a small molecule inhibitor of pseudouridine synthase PUS7, preferentially targets glioblastoma stem cells (GSCs) compared to NSCs.
PUS7 inhibitor C17 potently inhibits growth of multiple GSC lines (PBT003, PBT707, PBT726, and PBT111) in range 4 nM to 400 nM (IC50=92.15 nM, PBT003 GSCs).
PUS7 inhibitor C17 displays no inhibitory effect on control brain-derived normal NSCs.
PUS7 inhibitor C17 could inhibit pseudouridine modification by PUS7 and suppress GSC growth.
PUS7 inhibitor C17 suppresses tumor progression, prolong the lifespan of tumor-bearing mice.
PUS7 modifies tRNAs, reduces TYK2 translation, and downregulates a proliferation-restricting interferon-STAT1 pathway in glioblastoma.
