POMHEX is a cell-permeable, small-molecule ENO2-specific enolase inhibitor, exhibits selective killing against ENO1-deleted glioma cells with IC50 of 30 nM.
POMHEX displays weak acitivty against non-target ENO1-WT cells (IC50>1.5 uM).
POMHEX selectively induces energy stress, inhibits proliferation, and triggers apoptosis in ENO1-deleted glioma cells.
POMHEX eradicates intracranial orthotopic ENO1-deleted tumors in mice at doses well-tolerated in non-human primates.
