PAR4 Agonist Peptide AYPGKF-NH2 (PAR4-AP) is a potent, PAR4-selective agonist peptide, stimulates platelet aggregation in vitro with EC50 of 15 uM, shows no significant interaction with either PAR1 or PAR2.
AYPGKF activates and desensitizes PAR4 in platelets and, like thrombin, triggers phosphoinositide hydrolysis but not inhibition of adenylyl cyclase in PAR4-expressing cells.
AYPGKF is more potent than the native GYPGKF sequence at stimulating PAR4-mediated phosphoinositide hydrolysis.
AYPGKF effectively desensitizes PAR4-mediated signaling in human platelets.
