Neoseptin-3 is a specific synthetic agonist of mouse toll-like receptor 4 (TLR4)/myeloid differentiation factor 2 (MD-2) complex, promotes macrophage TNFα production with EC50 of 18.5 uM.
Neoseptin-3 induces TNFα, IL-6, and IFN-β secretion in different mouse cells.
Neoseptin-3 does not require TLR2, TLR3, TLR6, TLR7, or TLR9 to induce TNFα.
Neoseptin-3 specifically engages and activates mouse TLR4/MD-2, but cannot activate human TLR4/MD-2 or mixed heterodimers..
