NUCC-201760 is a specific small molecules inhibitor of mTORC2 complex component Stress-activated protein kinase-interacting protein 1 (SIN1) with IC50 of <1.5 uM in p-AKT S473 flow cytometry assay, selectively targets mTORC2 but not mTORC1.
NUCC-201760 uniquely inhibits RAS activity and downstream MAPK signaling, disrupts the ability of RAS to switch from the GDP to GTP form which requires SIN1.
NUCC-201760 enhances responses to different FDA approved pharmacological agents in U937 cells.
NUCC-201760 alters post-translational modification, protein-protein interactions, and blocks mTORC2- and rapamycin sensitive (RS) mTORC1-mediated signaling.
NUCC-201760 potently inhibits the growth of colon cancer, melanoma, and leukemia cell lines.
