NU9056 (NU 9056) is a specific Tip60 (KAT5) histone acetyltransferase inhibitor with IC50 of <2 uM.
NU9056 displays 16.5-, 29- and >50-fold for selectivity for Tip60 over PCAF, p300 and GCN5, respectively.
NU9056 (0-40 uM) decreased levels of acetylated histone H4K16, H3K14 and H4K8, targets for Tip60-mediated acetylation in treated LNCaP cells, also decreased acetylated tubulin in LNCaP cells (24 µM NU9056, 24 hours).
NU9056 reduces PSA and p53 protein levels, reduces LNCaP cell survival by inducing apoptosis, inhibits Tip60 accumulation and ATM phosphorylation in response to ionising radiation.
