NSC 87877 is potent, selective protein tyrosine phosphatase SHP1 and SHP2 with IC50 of 318 and 355 nM, respectively, also inhibits dual-specificity phosphatase 26 (DUSP26).
NSC 87877 displays 5-fold selectivity over PTP1B, 20-fold over HePTP and no activity against DEP1, CD45 and LAR.
NSC 87877 also effectively inhibits DUSP26-mediated dephosphorylation of p38.
NSC 87877 inhibits EGF-induced activation of Shp2 PTP, Ras, and Erk1/2 in cell cultures, inhibits EGFRvIII oncogenic transformation in human glioblastoma cells.
