NSC74429 is a CNS-active, small molecule PHLPP inhibitor with IC50 of <5 uM, shows neuroprotective activity in primary rat cortical neurons in STS assay.
NSC74429 increased 24 h neuronal survival in both the glutamate (at 50 µM) and H2O2 injury assay (at 25 and 50 µM).
NSC74429 (1 mg/kg) exhibits hippocampal neuroprotection mouse model of severe traumatic brain injury (TBI), significantly increased CA1 neuronal survival.
