NSC45586 free base (NCS 45586, NCI45586) is a potent, specific PHLPP2 inhibitor with IC50 of 4 uM, targets the PHLPP2 PP2C domain, suppresses MYC and triggers cell death.
NCI45586 caused increases in the phosphorylation of the downstream substrates GSK3 α/β on Ser21/9 and FoxO1/3α on Thr 24/32.
NCI45586 also induced a robust increase in the activity of Akt in HT29 cells (colon cancer cell line),
NCI45586 also showed suppression of MYC and some activation of AKT in the metastasis-derived DU145 cells.
