NSC194598 is a specific small molecule inhibitor that inhibits p53 sequence-specific DNA binding in vitro (IC50 = 180 nM) and in vivo.
NSC194598 selectively inhibits DNA binding by p53 and homologs p63/p73, but did not affect E2F1, TCF1, and c-Myc.
NSC194598 inhibits p53 DNA binding and induction of target genes.
NSC194598 apparently acts by binding to the DNA-binding domain of p53, and shows specificity against several control transcription factors.
NSC194598 suppresses p53 transcriptional output after DNA damage in culture and increases the survival of mice after irradiation.
