NS5806 (NS-5806) is a potent Kv4.3 channel activator that mediates the transient outward K+ current (Ito).
NS5806 increases IKv4.3 peak current amplitude in CHO-K1 cells expressing KV4.3 and KChIP2 with EC50 of 5.3 uM.
NS5806 also decays Kv4.1 independent of KChIP2, strongly inhibits Kv1.4 at 10 uM, but has no effect on Kv7.1, Kv11.1 and K(ir)2 currents.
NS5806 increases ventricular Ito with a leftward shift in Ito activation and accelerated restitution at 10 uM.
