NPD10084 (NPD 10084) is a novel compound that targets pyruvate kinase muscle isoform 2 (PKM2), exerts anti-proliferative activity against colorectal cancer cells in vitro (IC50=0.8-1.9 uM) and in vivo.
NPD10084 interrupted protein-protein interactions between PKM2 and β-catenin or STAT3, with subsequent suppression of downstream signaling, also inhibited the interaction between PKM2 and STAT3.
The anti-proliferative activity of NPD10084 may not be mediated by an alteration in the pyruvate kinase activity of PKM2.
NPD10084 partially suppressed the tumor growth of colorectal cancer cells in HCT116 xenograft mouse model.
