NITD-688 (EYU688) is a potent, specific pan-serotype inhibitor of dengue virus (DENV) NS4B protein, shows EC50 of 8-38 nM against all four DENV serotypes.
NITD-688 displays an EC50 value of 0.94 nM in PBMCs were infected with DENV-2.
NITD-688 shows potent activity against DENV (EC50 = 0.023 μM) in CPE assay, but not other flaviviruses or CHIKV.
NITD-688 directly binds to wild-type but not mutant NS4B with mutations T215A and A222V.
NITD-688 at 10, 30, or 100 mg/kg twice daily (BID) for 3 days or 30 mg/kg once daily (QD) for 3 days (oral gavage once daily (QD) or twice daily (BID) and was formulated in 0.5% methyl cellulose and 0.5% Tween 80 in water) reduces DENV-2 viremia during acute or delayed treatment of infected AG129 mice.
