NGM1 is a small molecule NOS1AP modulator of directly binds NOS1AP (CAPON) with SPR KD of 11.9 uM, alters NOS1AP-NOS1 (Nitric oxide synthase, nNOS) proximity, reduces viability in U87 and U251 GBM cells with IC50 values of 12.7 and 17.3 uM, respectively, with no effect on normal human astrocyte viability.

NGM1 is a small molecule NOS1AP modulator of directly binds NOS1AP (CAPON) with SPR KD of 11.9 uM, alters NOS1AP-NOS1 (Nitric oxide synthase, nNOS) proximity, reduces viability in U87 and U251 GBM cells with IC50 values of 12.7 and 17.3 uM, respectively, with no effect on normal human astrocyte viability.
NGM1 produces a concentration-dependent reduction in the NOS1-NOS1AP NanoBRET signal in CHO-K1 cells co-expressing NanoLuciferase-tagged NOS1 (N-terminal) and Venus-tagged CAPON (C-terminal).
NGM1 produces a concentration-dependent reduction in viability in both GBM models
NGM1 demonstrates reduced DNA synthesis, G0/G1 cell-cycle accumulation, and induction of apoptosis, accompanied by increased p53 and CDKN1A and decreased CDK6.
