NC1153 is a Mannich base compound, selective and orally active JAK3 inhibitor that blocks IL-2-induced activation of JAK3 and downstream substrates STAT5a/b.
NC1153 fails to inhibit several other enzymes, including growth factor receptor tyrosine kinases, Src family members, and serine/threonine protein kinases.
NC1153 inhibits proliferation of normal human T cells challenged with IL-2, IL-4, or IL-7.
NC1153 significantly extends survival of MHC/non-MHC mismatched rat kidney allografts, induces transplantation tolerance in vivo.
