Mpro61 is is a potent, selective, noncovalent and nonpeptidomimetic inhibitor of SARS-CoV-2 Mpro with Ki of 72 nM, 83 nM and 92 nM for WT Mpro and A173V and A173V/L50F variants respectively.

Mpro61 displayed a comparable level of inhibition against Mpro A173V and A173V/L50F with a similar Ki to the wild type, but not Nirmatrelvir and GC376.

Mpro61 failed to inhibits E166V/L50F double variant.
