MitoCDNB is a mitochondria-targeted molecule that disrupts thiol redox state and inhibits TrxR2, selectively disrupts mitochondrial thiol redox state in cells and in vivo, selectively depletes the mitochondrial GSH pool.
MitoCDNB reaction with GSH catalyzed by GST to form MitoGSDNB and deplete GSH, inhibits TrxR2 by alkylation of its active site selenol.
MitoCDNB inhibits recombinant mammalian cytosolic TrxR1 and mitochondrial TrxR2, MitoCDNB also alkylated Trx1 and Prx1, consistent with inhibition of these proteins by alkylation of their reactive thiols.
MitoCDNB is taken up rapidly by mitochondria and there converted to MitoGSDNB.
MitoCDNB enables assessment of the biomedical importance of mitochondrial thiol homeostasis in reactive oxygen species production, organelle dynamics, redox signaling, and cell death in cells and in vivo.
