MYF-03-176 is a potent and orally bioavailable MYF-03-69 analog, pan-TEAD inhibitor that disrupts YAP-TEAD association, suppresses TEAD transcriptional activity (IC50=11 nM).
Similar to MYF-03-69, MYF-03-176 led to a significant downregulation of YAP target genes CTGF, CYR61, ANKRD1 and an upregulation of BMF.
MYF-03-176 exhibits antiproliferative effect in NCI-H226 cells with IC50 of 9 nM.
MYF-03-176 potently inhibited Hippo signaling defective MPM cells and suppresses tumor growth in mesothelioma xenograft mouse model.
