MRK-1 is a small molecule non-nucleoside inhibitor of the RSV L polymerase protein, binds to the RNA-dependent RNA polymerase (RdRp, L-polymerase) and inhibits vRNA synthesis initiation.
MRK-1 exhibits antiviral activity in cell-based viral replication assays with antiviral potency against RSV-A2/EGFP (EC50 = 2.1nM) in a GFP reporter assay, the lab-adapted strains RSV-A Long (EC50 = 3.9nM) and 74 RSV-B Washington (EC50 = 3.3nM) in plaque reduction assays (PRA), and the clinical isolates RSV-A (EC50 = 4.3nM) and RSV-B (EC50 = 1.3nM) in cytopathic effect assays.
