MA-0204 (MA0204) is a potent, selective PPARδ modulator with EC50 of 0.4 nM (human PPARδ), dispalys no activity (10,000-fold selectivity) against PPARα and PPARγ.
MA-0204 exhibits potency for mouse PPARδ and rat PPARδ with EC50 of 7.9 nM and 10 nM, respectively.
MA-0204 also displays high selectivity (EC50 >10 µM) over other nuclear receptors,eg, androgen, progesterone and glucocorticoid receptors.
MA-0204 increases PPARδ target genes ANGPTL4 (angiopoietin like protein 4), CPT1a (carnitine palmytoyltransferase 1a) and PDK4, and improves fatty acid oxidation in vivo in mice and in vitro in patient-derived muscle myoblasts.
