LY3509754 is a potent, selective and oral small molecule inhibitor of IL-17A with high binding potency(KD=2.14 nM), dose-dependently prevents human IL-17A and IL-17AF from binding human IL-17RA.
LY3509754 displays no inhibition of IL-17C, IL-17E, or IL-17F interacting with their cognate receptors. LY3509754 dose-dependently inhibits IL-17A-induced CXCL1/GROα (IL-8 for canine) in primary or cell lines from species of interest (in human keratinocytes, IC50=8.25 nM; plasma protein binding-adjusted geomean, IC50=3.67 nM).
LY3509754 shows potential effect for treatment of psoriasis and other immune-mediated diseases.
