LUF7244 is a negative allosteric modulator of dofetilide binding to the Kv11.1 (hERG) channel with the strongest effect at 10 uM (IC50=3.9 uM).
LUF7244 significantly decreases the Kv11.1 affinities of typical blockers (ie, dofetilide, astemizole, sertindole, and cisapride).
LUF7244 prevents astemizole or sertindole caused heterogeneous prolongation of action potential duration in neonatal rat ventricular myocyte, with no effect on their viability, excitability and contractility.
