LPHN2 inhibitor D11 is a reversible and selective inhibitor of mechanosensitive GPCR latrophilin 2 (LPHN2, ADGRL2), inhibits mechanosensitivity of LPHN2 in HEK293 cells with IC50 of 20 nM.
D11 inhibited 3 pN force-induced LPHN2 activation in LPHN2-overexpressing HEK293 cells with IC50 of 39.88 nM.
D11 has no detectable effects on either the force-induced or agonist-induced activation of other adhesion GPCRs in the heterologous HEK293 overexpression system.
D11 dose-dependently inhibited MET currents in utricular hair cells with EC50 of 25 nM.
D11 (50 nM) abrogated the residual normal-polarity MET current in the BAPTA-treated utricular hair cells derived from the Pou4f3-CreER+/−;Lphn2+/+ mice.
