LPD-01 is a small molecule inhibitor of the Wnt/β-catenin signaling pathway, directly binds to importin7, inhibits the transactivation of target genes of the Wnt/β-catenin signaling pathway.
LPD-01 inhibits the Wnt/β-catenin signaling pathway without affecting β-catenin.
Importin7 is a member of the importin-βfamily of proteins of nuclear transport receptors that can act autonomously or in association with importin-β1.
LPD-01 inhibits the cell growth of human colorectal cancer cells with IC50 of 10.3 uM and 27.24 uM for HT-29 and SW480 cells, respectively.
LPD-01 reduces the expression of the target genes of the Wnt/β-catenin signaling pathway.
LPD-01 inhibits the nuclear translocation of importin7 cargoes such as Smad2 and Smad3 in TGF-β-stimulated HT-29 cells, and the knockdown of importin7 reduces the expression of the target genes of the Wnt/β-catenin signaling pathway.
