LC-YD03 is a potent, selective YTH domain-containing protein 1 (YTHDC1) inhibitor with IC50 of 41.98 nM and BLI Kd of 102 nM, shows little to no activity against YTHDF1, YTHDF2, YTHDF3, and YTHDC2.
LC-YD03 also shows no apparent off-target activity against METTL3/METTL14 or the tested kinase panel.
LC-YD03 retains activity against YTHDC1(WT), YTHDC1(N363A), and YTHDC1(S378A), with IC50 values of 290.7 nM, 396.9 nM, and 476.3 nM, respectively.
