JWX-A0108 is a novel selective type I α7 nAChR positive allosteric modulator (PAM) with EC50 of 4.35 uM, shows no potentiation effect on α3β4 nAChR, α4β2 nAChR, or 5-HT3AR.
JWX-A0108 selectively enhances the α7 current functionally expressed in oocytes.
JWX-A0108 (10 uM) potentiates the endogenous α7 current of cultured rat hippocampal neurons.
JWX-A0108 (10 uM) enhances GABAergic synaptic transmission in hippocampal brain slices.
JWX-A0108 (1-10 mg/kg, i.p.) reverses MK-801-induced deficits in auditory gating in MK-801-induced deficit in PPI in mice.
