JPC0661 is a specific small molecule direct inhibitor of AP1 transcription factor ΔdeltaFOSB (ΔFOSB), inhibits FOSB/JUND and ΔFOSB DNA-binding biochemically with IC50 of 9 uM and 52 uM respectively.
JPC0661 inhibits ΔFOSB-mediated transcription in cell-based assays with IC50 of 0.2 uM in luciferase reporter assays.
JPC0661 binds to a groove outside of the DNA-binding cleft of the ΔFOSB/JUND bZIP heterodimer.
JPC0661 disrupts DNA-binding directly, inhibits the ability of ΔFOSB to promote gene expression in cultured cells.
JPC0661 directly disrupts ΔFOSB–DNA interactions in vivo, inhibits ΔFOSB binding to AP1 consensus DNA-binding sites in vivo in an APP mouse model of AD neuropathology.
