JIB-04 is a pan-Jumonji histone demethylase inhibitor (IC50=230-1100 nM) with epigenetic activity, also potently inhibits SARS-CoV-2 replication in Vero E6 cells with EC50 of 695 nM.
JARID1A (KDM5A) is most sensitive to JIB-04 (IC50=230 nM), shows no inhibition of the mechanistically related TET enzymes.
JIB-04 specifically inhibits the activity of the Jumonji family of histone demethylases in vitro, in cancer cells, and in tumours in vivo.
JIB-04 showed in vitro antiviral activity in multiple cell types, including primary human bronchial epithelial cells, against several DNA and RNA viruses, including porcine coronavirus transmissible gastroenteritis virus.
