IsoLiPro is a novel small molecule USP11 inhibitor, reduces the USP11 protein level and suppressesthe USP11-mediated substrate deubiquitination.
IsoLiPro markedly reduces levels of total and phosphorylated tau in cultured cells overexpressing wild-type full-length human tau.
IsoLiPro markedly decreases USP11 protein level in SH-SY5Y cells and the mice brain.
IsoLiPro showed a strong binding affinity to USP11 (MST KD of 4.3394E-05 M), also demonstrated some extent of binding to other USPs, but the binding affinities are lower than USP11.
IsoLiPro markedly increases tau ubiquitination levels in HEK293-hTau cells.
IsoLiPro markedly induces tau clearance in the brain of 3xTg-AD and 5xFAD mice.
