IXA6 is a specific activator of IRE1/XBP1s signalling activator, promotes IRE1-dependent XBP1 splicing, selective activates the IRE1/XBP1s signaling.
IXA6 moderately increased IRE1-dependent Xbp1 splicing in wild-type mouse embryonic fibroblast (MEF) cells, but not in Ire1-deficient MEF cells.
IXA6 does not significantly activate ATF6 transcriptional signaling and PERK geneset.
IXA6 improves ER proteostasis of destabilized variants of amyloid precursor protein (APP) through an IRE1-dependent mechanism and reduces APP-associated mitochondrial toxicity in cellular models.
