IPI-443 is a potent, selective, orally active p110δ/γ PI3K dual inhibitor with cellular IC50 of 0.29 and 7.1 nM, respectively.
IPI-443 displays >600-fold selectivity over p110α and p110β.
IPI-443 potently reduces mouse B cell proliferation, survival, and plasmablast differentiation while increasing antibody class switching to IgG1。
IPI-443 potently inhibits human B cell proliferation in vitro at 10 nM, but not the p110γ selective inhibitor AS-252424.
