INI-43 (Inhibitor of Nuclear Import-43, JRMS) is a small molecule that bind nuclear import receptor Karyopherin beta 1 (Kpnβ1), inhibits Kpnβ1-mediated nuclear import with anti-cancer activity.
INI-43 significantly diminished NFAT activation in a dosedependent manner (IC50=9.3 uM in HeLa cells).
INI-43 prevents transcription factor and Kpnβ1 access to the nucleus, and INI-43 treatment results in cancer cell death via a G2/M cell cycle arrest.
INI-43 treatment induces cancer cell cycle changes and apoptosis and inhibits tumour growth in vivo.
