IMT1 (LDC195943) is a first-in-class specific inhibitor of mitochondrial transcription that target the human mitochondrial RNA polymerase (POLRMT).
IMT1 binds to an allosteric binding site near the active centre cleft of POLRMT.
IMT1 efficiently impairs mtDNA transcription in a reconstituted recombinant system and cause a dose-dependent inhibition of mtDNA expression and OXPHOS in cell lines.
IMT1 demonstrates anti-tumour effects against the growth of cancer cells and the persistence of therapy-resistant cancer stem cells.
