ID3-77 is a potent, covalent inhibitor of SARS-CoV-2 papain-like protease (PLpro) with IC50 of 209 nM, produces strong antiviral effect, reducing viral titer by 5.48 and 5.50 log units at 1 and 10 uM, respectively.
ID3-77 covalently labels catalytic Cys111 of PLpro.
ID3-77 shows selectivity over the cysteine proteases cathepsin B and calpain 1.
ID3-77 reduces SARS-CoV-2 lung viral load in vivo.
