HC-070 is a potent, selective, orally active inhibitor of TRPC4 and TRPC5 channel with IC50 of 9.3 and 46 nM (calcium influx inhibition), respectively.
HC-070 displays >400-fold selectivity over a wide range of ion channels, receptors, and kinases.
HC-070 inhibits lanthanum-activated hTRPC5-mediated currents with an IC50 of 0.52 nM in whole-cell manual patch clamp assay, also inhibits TRPC1/5 and TRPC1/4 heteromultimers with similar potency and reduces responses evoked by cholecystokinin tetrapeptide (CCK-4) in the amygdala.
HC-070 attenuates the anxiogenic effect of CCK-4 in the elevated plus maze (EPM), and shows anxiolytic and anti-depressant effects in vivo.
