GluN2B NAM 12 (GluN2B negative allosteric modulator 12) is a selective negative allosteric modulator of GluN2B NMDAR with IC50 of 31 nM and Ki of 15 nM (hGluN2B).
GluN2B NAM 12 also show high potency against GluN2B rat with Ki of 16 nM.
GluN2B NAM 12 displays no significant activity against GluN2- A/C/D (IC50 >10 uM).
GluN2B NAM 12 shows robust target engagement in rat with an ED70 of 1.4 mg/kg.
