GSK2556286 (GSK286) is a novel inhibitor of M. tuberculosis extracellularly in the presence of cholesterol and within human macrophages, inhibits growth within human macrophages with IC50 of 70 nM (H37Rv in THP-1 cells).
GSK2556286 (GSK286) exhibits no cross-resistance with known antitubercular drugs.
GSK2556286 (GSK286) shows efficacy in different mouse models of tuberculosis (TB) and has an adequate safety profile in preclinical species.
GSK2556286 (GSK286) is effective against both multidrug or extensively-resistant (M/XDR) and drug-sensitive (DS) M. tuberculosis with the potential to shorten the duration of treatment in novel combination drug regimens.
