GSK-843 (GSK'843) is a specific small molecule receptor-interacting protein kinase 3 (RIPK3) inhibitor, binds to RIP3 kinase domain with high affinity (IC50=8.6 nM).
GSK-843 (GSK'843) does not inhibit RIP1 kinase (RIPK1), and a panel of 300 human protein kinases.
GSK-843 (GSK'843) blocked TNF-induced necroptosis in a concentration-dependent manner in primary human neutrophils isolated from whole blood.
GSK-843 (GSK'843) inhibited TLR3- or DAI-induced death, two RIP1-independent pathways of necroptosis.
GSK-843 (GSK'843) induced caspase activation followed by apoptotic cell death in different cell types.
