GS-462808 is a potent Late INa current inhibitor of the cardiac Nav1.5 channel with IC50 of 1.33 uM.
GS-462808 has lower brain penetration and serendipitously lower activity at the brain isoforms, and improved CNS window (>20 rat and dog) relative to GS-458967.
GS-462808 shows improved anti-ischemic potency relative to ranolazine.
