Farudodstat (ASLAN003) is a highly potent, selective dihydroorotate dehydrogenase (DHODH) inhibitor with IC50 of 35 nM (human DHODH), significantly inhibits protein synthesis via activation AP-1 transcription, induces cell differentiation of AML cell lines.
Farudodstat (ASLAN003) inhibit leukemic cell proliferation of THP-1, MOLM-14 and KG-1 with IC50 values of 152 nM, 582 nM, and 382 nM, respectively.
Farudodstat (ASLAN003) decreases viability and induces differentiation in primary AML blasts and myelodysplastic syndrome samples, triggers apoptotic pathways via both mitochondrial and death receptor pathways.
Farudodstat (ASLAN003) (50 mg/kg, once daily oral gavage) demonstrates robust in vivo efficacy in multiple mouse xenograft AMLs.
