FTO-04 is a potent, selective, competitive small molecule inhibitor of m6A-RNA demethylase FTO with IC50 of 3.39 uM, 13-fold selectivity over RNA demethylase ALKBH5 (ALKBH5, IC50=39.4 uM).
FTO-04 prevented neurosphere formation in patient-derived glioblastoma stem cells (GSCs) without inhibiting the growth of healthy neural stem cell-derived neurospheres.
FTO-04 increased m6A and m6Am levels in GSCs consistent with FTO inhibition.
