FS2921 is a potential, functionally selective GluN1/GluN2A NMDAR positive allosteric modulator (PAM), selective versus GluN1/GluN2B and GluN1/GluN2D, increases NMDAR-mediated currents with EC50 of 8.8 uM.
FS2921 exhibits a weak NAM effect for GluN1/GluN2C but does not show any influence on native AMPA receptors and GABA receptors.
FS2921 slightly increases NMDA potency from 23.9 ± 1.3 to 13.6 ± 1.1 μM, but has no influence on the maximum currents induced by saturated NMDA.
FS2921 moderately decreased the immobility time in the FST.
FS2921 moderately reversed the reduction in social interactions induced by CSDS and exhibited an equal antidepressant effect compared with that of fluoxetine.
