FK3453 (FK 3453) is a novel inhibitor of thymic stromal lymphopoietin (TSLP) production, reduces both TSLP protein levels and mRNA levels both in vitro and in vivo.
FK3453 inhibits TSLP mRNA stimulated TNF-α (100 ng/mL) or TPA (30 nM) in PAM212 cells, but not TNF-α-induced TNF-α mRNA expression.
FK3453 inhibited TPA-induced the expression of IL-33 mRNA in NHEKs, did not affect TPA-induced phosphorylation of ERK.
FK3453 (one milliliter of 2% (w/v) CMC-Na in saline including LPS (10 ng/mL) with FK3453 (0.3 mM)) inhibited TSLP production in air-pouch type models in mice.
FK3453 is a lead compound for a novel type of medicine which prevents the onset and aggravation of allergic diseases.
